What is Gavreto?

Gavreto (pralsetinib) is an oral prescription medicine that may be used to treat the following cancers caused by abnormal RET (rearranged during transfection) genes that have spread (metastasized):

  • non-small cell lung cancer (NSCLC) in adults
  • advanced thyroid cancer or thyroid cancer in adults and children 12 years of age and older who require a medicine by mouth or injection (systemic therapy) and who have received radioactive iodine and it did not work or is no longer working.

âś… Indications:

GAVRETO 100 mg is used to treat cancers with RET gene alterations, including:

1. RET Fusion–Positive Non-Small Cell Lung Cancer (NSCLC)

  • Advanced/metastatic

  • First-line or second-line treatment

2. RET-Mutant Medullary Thyroid Cancer (MTC)

  • Locally advanced or metastatic

  • For patients who require systemic therapy

3. RET Fusion–Positive Thyroid Cancer

  • Radioactive iodine–refractory (if eligible)

âś… RET mutation or fusion must be confirmed with a validated molecular test (e.g. NGS, FISH).


đź’Š Dosage (for Adults):

  • 400 mg once daily (4 x 100 mg capsules)

  • Take on an empty stomach (at least 2 hours before or after food)

  • Continue until disease progression or toxicity


⚙️ Mechanism of Action:

GAVRETO is a selective RET kinase inhibitor
It blocks the RET signaling pathway, which is abnormally activated in certain cancers due to RET mutations or gene fusions, helping to shrink or slow tumor growth.
