🧬 Indications

 

Ayvakit is a prescription medicine used to treat adults with certain types of gastrointestinal stromal tumor (GIST) and systemic mastocytosis (SM). Ayvakit contains a drug called avapritinib, which belongs to the class of medicines called tyrosine kinase inhibitors. GIST is a cancer of the gastrointestinal tract most often caused by mutations in the platelet-derived growth factor receptor alpha (PDGFRA) protein, which is part of a family of proteins called receptor tyrosine kinases (RTKs). Ayvakit is used to treat:

  1. Advanced Systemic Mastocytosis (AdvSM)

  2. Aggressive Systemic Mastocytosis (ASM)

  3. Systemic Mastocytosis with Associated Hematologic Neoplasm (SM-AHN)

  4. Unresectable or metastatic gastrointestinal stromal tumor (GIST) with PDGFRA exon 18 mutations, including PDGFRA D842V mutation

        Approved by the US FDA for these rare diseases where other treatments are often ineffective.

🧪 Typical Dosage

 

  • For PDGFRA exon 18 mutant GIST:
    ✅ 300 mg once daily

  • For advanced systemic mastocytosis:
    ✅ Start with 200 mg once daily
    ✅ May reduce based on toleran

  • Always taken on an empty stomach — 1 hour before or 2 hours after a meal.


⚠️ Side Effects

     Common:

 

  • Edema (swelling)

  • Nausea, vomiting, diarrhea

  • Fatigue, abdominal pain

  • Cognitive effects (memory impairment, confusion)

  • Low blood counts (anemia, thrombocytopenia)

     Serious:

 

  • Intracranial bleeding (rare)

  • Cognitive impairment (dose-related; reversible after stopping or reducing dose)


🚫 Warnings and Precautions

 

  • Avoid strong CYP3A inhibitors/inducers

  • Caution in patients with CNS conditions

  • Baseline brain imaging may be recommended in patients with neurologic symptoms
