What is Cibinqo?

Cibinqo (abrocitinib) tablets are used to treat eczema (atopic dermatitis) that is moderate to severe in patients who have not found relief with topical medications or are not able to use them. Cibinqo is a once-a-day tablet that can be used by adults and children over 12 years of age. Eczema may be caused by an overactive immune system leading to inflammation which causes dry, itchy skin and eczema flare-ups. Janus kinase (JAK) is an enzyme thought to play a role in skin inflammation. Cibinqo works by blocking the JAK1 activity, decreasing inflammation which helps reduce itching and improves skin condition.

CIBINQO® (abrocitinib) is an oral medication approved for the treatment of moderate-to-severe atopic dermatitis (eczema) in adults and adolescents aged 12 years and older. It is particularly indicated for patients whose condition is not adequately controlled with other systemic therapies, including biologics, or when such treatments are inadvisable.

🔬 Mechanism of Action

CIBINQO is a selective Janus kinase 1 (JAK1) inhibitor. By inhibiting JAK1, it disrupts the signaling pathways of various pro-inflammatory cytokines involved in the pathogenesis of atopic dermatitis, leading to reduced inflammation and alleviation of symptoms such as itching.


💊 Dosage and Administration

  • Recommended Starting Dose: 100 mg taken orally once daily.

  • Dose Adjustment: If the response is inadequate after 12 weeks, the dose may be increased to 200 mg once daily.

  • Dose Reduction: In patients with moderate renal impairment or when co-administered with strong CYP2C19 inhibitors, a starting dose of 50 mg once daily is recommended.

  • Administration: CIBINQO can be taken with or without food.

Clinical trials have demonstrated that CIBINQO provides significant improvements in skin clearance and itch relief

  • At 12 weeks, 41% of patients taking 200 mg and 26% taking 100 mg achieved clear or almost clear skin, compared to 8% with placebo.

  • Rapid itch reduction was observed as early as week 2 in some patients.

  • Thrombosis (blood clots)

  • Laboratory abnormalities (e.g., thrombocytopenia, lymphopenia, lipid elevations)

Monitoring:

  • Prior to initiating therapy, patients should be evaluated for tuberculosis and other infections.

  • Regular monitoring of blood counts, liver enzymes, and lipid profiles is recommended during treatment.
