ā Indication
MitotaneĀ is used to treatĀ cancerĀ of theĀ adrenal glands. It works by slowing the growth of or killing adrenal gland cells and also decreases the amount of hormones made by the adrenal gland. Used to treat inoperable, metastatic, or recurrent adrenocortical carcinoma (ACC) ā a rare cancer of the adrenal gland.
āļø Mechanism of Action
Mitotane works by destroying adrenal cortex cells, thereby reducing the production of adrenal hormones.
It has cytotoxic and adrenolytic effects.
š§¾ Dosing (Typical Starting Dose)
2ā6 grams per day, divided into 2ā3 doses (dose is adjusted based on tolerance and blood levels).
Therapeutic drug monitoring is required to maintain effective mitotane plasma levels.

Nausea, vomiting
Diarrhea
Fatigue
Dizziness or confusion
Weight loss
Adrenal insufficiency (requires steroid replacement)
Warnings
You may need to take steroid medicine, or stop taking Lysodren for a short time if you have a serious injury or medical emergency. Your doctor will determine when you can start taking Lysodren again.Ā Do not change your doses or medication schedule without advice from your doctor.
You must remain under the care of a doctor while taking Lysodren.

Before using thisĀ medication, tell your doctor or pharmacist your medical history, especially of:Ā liver disease,Ā kidney disease, bleeding problems.
Mitotane can make you more likely to get infections or may make current infections worse. Stay away from anyone who has an infection that may easily spread (such asĀ chickenpox, COVID-19,Ā measles,Ā flu). Talk to your doctor if you have been exposed to an infection or for more details.
